New antibodies against 2 ectonucleotidases

Ectonucleotidases are enzymes that break down ATP on the extracellular surface. They have several important physiological functions, e.g. they help to control the immune response or influence purinergic signal transmission.
We have introduced a new rat monoclonal antibody against mouse CD39, also known as ENTPD1 (ecto-nucleoside triphosphate diphosphohydrolase). This enzyme that catalyzes the transition from ATP and ADP into AMP. CD39 is highly expressed on vascular and immune cell populations such as B cells, NK cells, dendritic cells, Langerhans cells, monocytes, macrophages, mesangial cells, neutrophils, regulatory T cells and microglia. In the brain this protein is involved in the branching of microglial processes and blood-brain barrier function. In microglia, up-regulation of CD39 expression has been shown in several neuroinflammation models and associated with a switch from an inflammatory to an anti-inflammatory phenotype.

Indirect immunostaining of a formalin fixed paraffin embedded section of mouse kidney with anti-mouse CD39 antibody (cat. no. HS-493 017, dilution 1:100, DAB, brown). Nuclei have been counterstained with haematoxylin (blue).

The second new antibody is directed against mouse CD73, also known as NT5E (ecto-5′-nucleotidase). This cell surface ectoenzyme is involved in the conversion of AMP (from the enzymatic action of CD39) to adenosine. CD73 is expressed by several cell types, including endothelial cells, epithelial cells, B- and T-cells. In the latter, CD73 is mainly expressed by regulatory T cells. As regulators of the dynamic balance between immune-activating extracellular ATP and immunosuppressive adenosine, CD39 and CD73 impair the ability of immune cells to exert anticancer immunity and play an important role in the immune escape of tumor cells within the tumor microenvironment.

Indirect immunostaining of a formalin fixed paraffin embedded section of mouse lung with rabbit anti-CD73 antibody (cat. no. HS-492 003, dilution 1:1000, DAB, brown). Nuclei have been visualized by haematoxylin staining (blue).

New recombinant rabbit monoclonal antibody against Homer 1b/c

Homer 1 is a scaffolding protein of the post synaptic density (PSD) and enriched at excitatory synapses. The protein binds metabotropic glutamate receptors, TRPC1, proteins of the Shank family and others. By aggregating these proteins into clusters, Homer 1 was suggested to organize distinct signaling domains in the PSD. This antibody is specific for Homer 1b/c and displays no cross-reactivity against the Homer 1a isoform.

Indirect immunostaining of a formaldehyde fixed rat hippocampus section (dentate gyrus) with rabbit anti-Homer1b/c antibody (cat.. no. 160 018, dilution 1:500, red) and guinea pig anti-NeuN antibody (cat. no. 266 004, dilution 1:500, green). Nuclei have been visualized by DAPI staining (blue).
