Polyclonal guinea pig anti-Pyruvate decarboxylase (PC) antibody

Pyruvate carboxylase is a mitochondrial enzyme catalyzing the ATP-dependent carboxylation of pyruvate to oxaloacetate. It plays an essential role in controlling whole-body energetics through regulation of gluconeogenesis. PC shows high expression in liver and kidney, the primary organs of gluconeogenesis. In the brain, PC replenishes the intermediates of the tricarboxylic acid cycle and has an important anaplerotic function. It is predominately expressed in astrocytes and is needed for the de novo synthesis of glutamate, the main excitatory neurotransmitter (see Featured Topic The Glutamatergic Synapse). Recent literature indicates that PC is also expressed in a subpopulation of neurons.

This knock-down validated new antibody is the perfect tool to analyze the important metabolic "control point" pyruvate carboxylase in brain.

Indirect immunostaining of formaldehyde fixed rat hippocampus mixed cultures with guinea pig anti-Pyruvate carboxylase antibody (cat. no. 494 005, dilution 1:500, red), rabbit anti-MAP2 antibody (cat. no. 188 002, dilution 1:1000, green) and mouse anti-GFAP (cat. no. 173 011, dilution 1:1000, blue). Nuclei have been visualized by DAPI staining (white).

New antibody against Dopamine β-hydroxylase (DBH)

Dopamine β-hydroxylase (DBH), also referred to as dopamine β-monooxygenase, is an enzyme that catalyzes the conversion of dopamine to noradrenaline/norepinephrine and of p-Tyramine to p-Octamine. Norepinephrine (NE), also termed noradrenaline, is a neurotransmitter that makes the body “ready for action” often referred to as the fight or flight hormone. DBH is expressed in noradrenergic neurons of the central nervous system (e.g.locus coeruleus), the peripheral nervous system (e.g. sympathetic ganglia) and by chromaffin cells in large dense core vesicles (LDCVs) of the adrenal medulla.

Together with antibodies against NET (norepinephrine transporter), this new guinea polyclonal anti-DBH antibody is a good marker for noradrenergic neurons.

Indirect immunostaining of a formaldehyde fixed mouse adrenal gland (medulla) section with guinea pig anti-Dopamine β-hydroxylase antibody (cat. no. 496 004, dilution 1:1000, red) and rabbit anti-ChromograninB antibody (cat. no. 259 103, dilution 1:500, green). Nuclei have been visualized by DAPI staining (blue).

Investigate neuroinflammation and blood-brain-barrier damage with the new CD169 antibody

CD169, also known as Siglec-1 or sialoadhesin, is a cell surface receptor that is most frequently expressed by certain macrophage subsets in lymphoid tissue. CD169+ macrophages are involved in immunological tolerance, antigen presentation and defense against infectious agents such as viruses and play a tumor-suppressive role in malignant tumors. In the intact brain, CD169 stains subpopulations of macrophages in the choroid plexus, leptomeninges and circumventricular organs. CD169 is regulated by contact with plasma proteins, and damage to the blood-brain-barrier leads to the expression of CD169 on microglia and macrophages within the parenchyma.
The new rat monoclonal antibody against murine CD169 (no signal in human or rat tissues) works superior in IHC and FFPE (IHC-P) applications.

Indirect immunostaining of a formalin fixed paraffin embedded mouse spleen section using Tyramide signal amplification with rat anti-CD169 (cat. no. HS-495 017, dilution 1:500, red) and rabbit anti-F4/80 (cat. no. HS-397 008, dilution 1:500, green). Nuclei have been visualized by DAPI staining (blue).
