Introduction
The acetylene group, recognized for its unique chemical properties, is extensively utilized in bioorthogonal chemistry and medicinal chemistry. It plays a key role in copper-catalyzed azide-alkyne cycloaddition (CuAAC) and strain-promoted azide-alkyne cycloaddition (SPAAC), enabling selective and bioorthogonal reactions within complex biological environments. These reactions are crucial for the development of imaging and drug delivery. Furthermore, there are many roles that this structural feature can play in drug activity, which include potency enhancement, reactive warhead, nonpolar linear rigid spacer, bioisostere to a wide range of functionalities, and modulation of drug metabolism pharmacokinetic (DMPK) profile. So far, Chemscene has developed many acetylene group-containing building blocks.
