Accutase®는 천연 효소 혼합물로서 고착 세포를 세포배양 접시에서 떼거나 조직으로부터 primary cell들을 분리할 때 사용할 수 있습니다. Accutase® 대부분의 세포에 사용할 수 있으며 사람 중간엽줄기세포 또는 사람 신경줄기세포에도 적용할 수 있습니다.
Accutase® is a natural enzyme mixture for detachment of adherent cells or dissociation of primary cells from tissues.
Accutase® can be used for most cells, even human mesenchymal stem cell or human neural stem cell.

Tested Cell Lines of Accutase®
hESCs, fibroblasts, keratinocytes, vascular endothelial cells, hepatocytes, vascular smooth muscle cells,
hepatocyte progenitors, primary chick embryo neuronal cells, bone marrow stem cells, adherent CHO and BHK cells,
macrophages, 293 cells, L929 cells, immortalized mouse testicular germ cells, 3T3, Vero, COS, HeLa, NT2,
MG63, M24 and A375 metastatic melanoma, gliomas U251 and D54, HT1080 fibrosarcoma cells, and Sf9 insect cells.
"Always Gentle" reattachment of neuronal stem cells passaged with Accutase.
Human neural stem cells freshly dispersed by Accutase seeded on the glass coverslip bottom of a WPI Fluorodish,
timelapse-imaged over a period of 6 days.

Human MG63 Fibrosarcoma cells cultured in DMEM + 10% FBS were treated with Accutase. Cell viability was measured by each different treated time. Treatment resulted in rapid cell detachment, a single cell suspension, and high viability. Accutase is gentle on cells; viability was 97 ± 3% even after 45 minutes in Accutase.

Human ES cells are treated with Accutase or Collagenase and cryopreserved. After stored in liquid nitrogen for two months and cultured, total cell numbers and live cells numbers are counted after a day. Accutase treated cells (green) has superior cell viability compared to collagenase treated cells.
Overview of Protocol
1. Carefully aspirate the medium from cultured dish/well/flask.
2. Add enough amount of Accutase to cover the cells.
3. Incubate for 5 to 10 minutes at room temperature (up to a maximum of 1 hour)
4. Check the cells frequently to see if they have rounded and no longer attached to the bottom of dish/well/flask.
5. Count the cells and passage. No need to add enzyme inhibitors or rinse.
