Introduction

Pyridazinone is a six-membered heterocyclic scaffold featuring two adjacent nitrogen atoms and a carbonyl group. The embedded hydrazine-like (N-N) motif contributes to a semi-rigid framework and serves as a versatile pharmacophore capable of forming strong hydrogen bonds with biological targets. Furthermore, the electron-withdrawing carbonyl group polarizes the ring, thereby enhancing target binding affinity by providing a key hydrogen bond from the N-H group. Moreover, the scaffold's synthetic versatility facilitates the optimization of its physicochemical and pharmacokinetic properties, such as solubility and metabolic stability. As a result of this multifaceted profile, Pyridazinone has been successfully utilized as a privileged structure in developing therapeutics for the treatment of cardiovascular, inflammatory, and oncological diseases.
