Antibody-drug conjugates (ADCs) represent an emerging cancer therapeutic strategy. To date, 19 ADCs have been approved worldwide. Among them, three incorporate PEG linkers to optimize stability, solubility, and payload delivery:

• Trodelvy® (Sacituzumab govitecan) – Conjugated to SN-38 via a cleavable maleimide linker with a short PEG unit.
• Zynlonta® (Loncastuximab tesirine) – Conjugated to a PBD dimer via a cleavable maleimide linker containing a PEG8 spacer.
• Jiataile® (Sacituzumab tirumotecan, China) – Conjugated to a belotecan-derived topoisomerase I inhibitor via a cleavable CL2A linker incorporating PEG8.

PEG linkers play a vital role in ADC performance by reducing aggregation, lowering immunogenicity, and extending circulation time. These features make PEG a key component in modern ADC design.
