Baseclick’s RNA Labeling Kit has been developed for the production of alkyne-labeled RNA by in vitro transcription (IVT) from bacteriophage T7 RNA polymerase promoters.
An alkyne-modified nucleotide, 5-Ethynyl-UTP (5-EUTP), is first incorporated into the RNA. Then, the EU-containing RNA is reacted with azido-derivates of fluorescent dyes, haptens and others (a classical click reaction).
The rate of label incorporation can be tuned by varying the concentration of 5-EUTP in the IVT reaction or by the amount of azide-containing reagent used in the click reaction. This allows to generate RNA batches with different labels from a single batch of EU-containing RNA, providing additional flexibility in mix and matching multiple probes/reporters. 

The labelled RNA can be used as probe for classical hybridisation-based applications (e.g., ISH/FISH, blots, etc.). In addition, the ability to finely tune the RNA labelling rate, allows generating labelled mRNA suitable for simultaneous expression of exogenous proteins and mRNA tracking upon transfection in cultured cells and living organisms (see Croce et al.).
